Lauryl Sulfate Broth (LSB)

Application: This selective culture medium is used for the selective identification and enrichment of coliforms in water, wastewater, milk, ice cream, dairy products, and other materials that are of sanitary importance.

Standards: It conforms to ISIRI 11166, ISO, APHA (Iran national standard).

  • Shake the container of lauryl sulfate broth well. Dissolve 35.6 grams of the medium in 1 liter of distilled water. Mix thoroughly.
  • Heat gently until the medium is completely dissolved in the water.
  • After the medium has dissolved, distribute it into test tubes containing Durham tubes.
  • Autoclave at 121 degrees Celsius for 15 minutes.
  • To prevent bubbling inside the Durham tubes, allow the autoclave to cool to at least 75 degrees Celsius before opening it.

STORAGE

The medium powder should be kept tightly closed and stored at temperatures below 30 degrees Celsius, and the medium should be stored at a temperature of 2-8 degrees Celsius. The best time to use is before the expiration date printed on the product label.

  • Inoculate the samples in a culture plate and incubate for 24-48 hours under aerobic conditions at a temperature of 35±2 degrees Celsius.

Results and Interpretation

  • If the cultivation is done on a plate, any blackening of the color of the cultivation medium in the plate indicates a positive reaction, and the lack of blackening indicates a negative reaction.
  • In cases where the sample has been inoculated in the form of a slant in the prepared cultivation medium, if more than half of the slant has turned black after 24-48 hours, the test result will be positive, and if less than half or none of it has turned black after this period, the test result will be negative.

Note

  • Strains of lactococcus, leuconostoc, and pediococcus bacteria isolated from human infections have had a positive reaction in this cultivation medium.
  • Rare strains of viridans streptococci are capable of blackening the bile esculin agar cultivation medium and creating a weak positive reaction.
